B.078EFFECTS OF THE ACUTE ADMINISTRATION OF ETHANOL AND ASTAXANTHIN ON THE PROPAGATION OF CORTICAL SPREADING DEPRESSION

Resumo

Objectives: We have previously demonstrated that chronic treatment (one daily dose during 18 days) with ethanol facilitated in adult rats the propagation of cortical spreading depression (CSD), and the carotenoid astaxanthin (Ax) antagonized such effect (Alcoholism: Clin.Exp.Res.32:1417, 2008). Here we investigated the effects of a single administration of ethanol combined or not with a single dose of Ax, on CSD propagation. Methods: Male Wistar adult rats (150-165 days of age; n=9) received per gavage Ax (10ug/kg, dissolved in olive oil). One hour later, a second gavage with 3g/kg ethanol was administered (group Ax+Etoh). Four additional groups were treated with the same protocol, with either water or oil instead of Ax in the first gavage (groups water+etoh and oil+etoh; n=9 for both groups), and replacing ethanol with water in the second gavage (groups Oil+Water; n=10 and Ax+Water; n=11). Immediately after the second gavage, under urethane+chloralose anesthesia, the CSD was recorded in two points of the cortical surface during 4h and the CSD velocity of propagation was calculated, based on the time spent for CSD to pass the interelectrode distance. Results: Ethanol treatment (groups Water+Etoh and Oil+Etoh) reduced (p<0.05; ANOVA plus Tukey test) the CSD velocity, as compared to the group Oil+Water (mean+sd CSD velocities, in mm/min, were respectively 2.75+0.31, 2.71+0.27 and 3.05+0.19). Ax alone did not effect CSD propagation (group Ax+H2O; 2.89+0.07), but prevented the CSD reduction associated to ethanol (group Ax+Etoh; 3.13+0.16). Conclusion: In contrast to the chronic treatment, acute ethanol administration antagonized CSD propagation. Astaxanthin, however, attenuated the ethanol action, reinforcing the role of this antioxidant molecule on brain processes involved in CSD.
